A basic wet mount places a thin, suitable specimen in a small drop of water under a coverslip. It is useful for temporary, non-medical observations with a compound microscope. If you want labeled examples without preparing a specimen, start with the prepared-versus-blank microscope slide guide.
Safety boundary: glass slides and coverslips can chip or cut. Use intact glass with appropriate supervision, handle it by the edges, and discard damaged pieces safely. This guide does not cover blood, saliva, wounds, cultures, unknown water samples, diagnosis, stains, chemicals, heat, needles, scalpels, or other sharps.
What you need
- One clean, undamaged blank microscope slide
- One clean, undamaged coverslip
- A dropper containing clean water
- Blunt tweezers if needed
- A small piece of absorbent paper
- A thin, clean, non-hazardous specimen such as a short fiber, a tiny paper fragment, or another material approved for the activity
How to make a simple wet mount
- Check the glass. Do not use a chipped, cracked, or dirty slide or coverslip. Handle both by their edges.
- Place the specimen. Put a very small, thin specimen near the center of the slide. A thick or opaque object is usually a poor fit for transmitted-light viewing.
- Add one small drop of water. Use only enough to surround the specimen without flooding the slide.
- Lower the coverslip. Rest one edge of the coverslip beside the drop, hold it at an angle, and lower it slowly. This reduces—but does not guarantee the absence of—air bubbles.
- Remove excess water. Touch absorbent paper to the edge of the coverslip so it wicks away excess liquid. Do not press down on the coverslip.
- Start at the lowest objective. Center the specimen and focus at low power before moving to a supported higher objective. Keep the objective from contacting the coverslip.
Why coverslip thickness and objective markings matter
Microscope objectives can be designed for a particular optical system, imaging medium, tube length, and cover-glass thickness. Nikon’s current objective-specification guide notes that 0.17 mm is the standard cover-glass thickness for most transmitted-light applications, while some objectives require a correction collar or no coverslip at all. Read the exact objective markings and the microscope manufacturer’s documentation rather than assuming every coverslip or objective is interchangeable.

After viewing
Remove the slide from the stage before cleaning the microscope. Follow the slide supplier’s handling instructions and the microscope manufacturer’s cleaning guidance. Thermo Fisher’s storage guidance calls for keeping slides and coverslips dry and avoiding large temperature and humidity changes. Do not reuse damaged glass.
